In both situations, if only the batter missed the respective bases then only his run would not count. Therefore the answer to each situation would be: 2 runs score.
Any appeal that takes place for a runner missing a base while rounding the base only affects that runner's run. So if it had been the first runner who missed 3rd and home and the appeal took place, only his run would be deducted and the subsequent two runs would not be affected.
If more than two outs then, it would depend on where the lead runners were when the runner who missed the bases missed 1st. The appeal at home would result in two runs scoring.

Depends on the number of outs already recorded.The batter is automatically out for missing any bases. This does not effect the base runners who can advance as much as they can.

With 2 outs - 0 runs score as the batter has to reach 1st base safely for the runs to score. If he never touched first it is the same as getting forced out at first.
With less than 2 outs - the batter is out, but the other two runs score.
