The first home run ever hit in the National League was by Ross Barnes of the Chicago White Stockings (now known as the Chicago White Sox), in 1876.

This is hard to pin point, because it depends on what you consider the start of MLB. In the late 1800's the different leagues were merging and failing, and it is really hard to pinpoint when baseball historians really don't have a definitive starting point.

Hell, some can't even agree on who invented baseball.
